ABSTRACT

Objective:To study the effects of arsenic exposure on necroptosis pathway and inflammatory response of mouse myocardial cells.Methods:Sixty male C57BL/6J mice were randomly divided into control group (group C) and low, medium, and high dose arsenic exposure groups (groups L, M, H) based on body weight using a random number table method. Each group had 15 mice, and they drank 0.00, 0.15, 1.50, and 15.00 mg/L arsenic trioxide (As 2O 3) solution prepared with deionized water. The exposure period was 12 weeks. Hematoxylin-eosin (HE) staining and Masson trichrome staining of paraffin-embedded heart tissues were used to observe the histopathology changes of the heart. Transmission electron microscopy (TEM) was used to evaluate the ultrastructural changes of myocardial cells. The quantitative real-time PCR (qRT-PCR) was used to detect the mRNA expression of inflammatory genes [tumor necrosis factor (TNF)-α and interleukin(IL)-6] and the genes involved in necroptosis pathway [receptor-interacting protein (RIP) 1, RIP3 and mixed-lineage kinase domain-like protein (MLKL)]. Protein expressions of RIP1 and RIP3 in the heart were assessed by western blotting. Results:Histopathological examination results showed there were myocardial necrosis, inflammatory cells infiltration and fibroblasts hyperplasia and other changes in groups M and H. TEM analysis revealed marked ultrastructural changes in groups M and H, including fractured myofibril, fractured Z lines of sarcomere, and swollen mitochondria with fractured cristae. Compared with group C (1.00 ± 0.00), the mRNA expression of RIP1 in group H was significantly up-regulated (1.41 ± 0.06, P < 0.05); the mRNA expressions of RIP3 (1.29 ± 0.14, 1.56 ± 0.08), MLKL (1.23 ± 0.05, 1.36 ± 0.07), TNF-α (2.20 ± 0.10, 2.23 ± 0.18) and IL-6 (1.87 ± 0.16, 1.63 ± 0.15) were significantly up-regulated in groups M and H ( P < 0.05). The protein expressions of RIP1 (0.43 ± 0.04, 0.50 ± 0.04) and RIP3 (0.68 ± 0.02, 0.84 ± 0.05) in groups M and H were higher than those in group C (0.25 ± 0.01, 0.45 ± 0.04, P < 0.05). Conclusion:Subchronic arsenic exposure induces histopathological changes such as myocardial necrosis and fibrosis in mice, inducing necroptosis and inflammatory reactions in myocardial cells.

ABSTRACT

OBJECTIVE:To provide scientifi c evidence for improving the quality standard of Mongolian medicine Juniperus rigida. METHODS :Totally 10 batches of J. rigida from different places were taken as samples to observe their characters and identify them by microscope ;TLC method was adopted to qualitatively identify isoquercitrin ,quercitrin,amentoflavone, podocarpusflavone A and hinokiflavone ;the contents of total ash ,acid-insoluble ash ,ethanol-soluble extract and heavy metals were determined by related method stated in 2020 edition of Chinese Pharmacopeia (part Ⅳ). The contents of above 5 components in samples were determined by HPLC. RESULTS :The powder of J. rigida was green or yellowish green ,polygonal tracheids , closely arranged in longitudinal with unequal stomatal ;epidermal cells were nearly rectangular ;sclerenchyma cells were quasi rectangular and the wall beadedly thickening. Results of TLC showed that the spots of the same color were found in the corresponding positions of chromatogram for test sample and substance control. The contents of total ash ,acid-insoluble ash and ethanol-soluble extract in 10 batches of samples were 7.37%-11.18%,0.75%-2.98%,16.55%-26.42%,respectively;average contents were 8.51%,1.27%,22.35%. The contents of lead ,arsenic,cadmium,mercury and copper were 2.00-5.44,0.65-1.65, 0.044-0.100,0.034-0.160,4.59-6.79 mg/kg,respectively;average conte nts were 3.73,0.97,0.078,0.061,5.23 mg/kg. The linear ranges of isoquercitrin ,quercitrin,amentoflavone,podocarpus- flavone A and hinokiflavone were 4.98-20.02,49.99-199.96, 19.94-99.96,9.99-40.00,20.20-159.98 μg/mL(all r>0.999 7); com RSDs of precision ,repeatability and stability (24 h) tests were all less than 3.00%(n=6);the average recoveries were 话:0993-2057878。E-mail:Tanghuishz@qq.com 100.62%-102.96%,RSDs were 1.21%-1.88%(n=6). Average contents of the above-mentioned 5 compounds in 10 batches of samples were 0.089-0.379,1.379-4.250,1.077-2.026,0.162-0.423, 0.016 9-0.117 0 mg/g,respectively. CONCLUSIONS :The qualitative and quantitative analysis methods of Mongolian medicine J. rigida are established. It is preliminarily proposed that the total ash content shall not exceed 10.22%,the acid-insoluble ash content shall not exceed 1.53%,ethanol-soluble extract content shall not be less than 17.88%,heavy metal lead should not exceed 5 mg/kg,arsenic should not exceed 2 mg/kg,cadmium should not exceed 0.3 mg/kg,mercury should not exceed 0.2 mg/kg,copper should not exceed 20 mg/kg.

ABSTRACT

OBJECTIVE:To improve the quality standard of M ongolian med icine Artemisia sacrorum ,and to provide scientific basis for comprehensive quality evaluation. METHODS :The appearance and microscopic characteristics of A. sacrorum were identified;scopoletin,chlorogenic acid ,caffeic acid ,scopoletin and 3,5-dicaffeoylquinic acid were identified quantitatively by TLC;the contents of above 5 components were determined by HPLC. The water content ,total ash and extract were examined. RESULTS:The stem of A. sacrorum was cylindrical ,and its surface was purple or purple-brown or cyan-brown ;the leaves were ovate or oblong-ovate ,fragrant;the flowers were yellow ,head-shaped,subglobose or hemispherical. The powder was green or yellow-green,its pollen grain had three germination ;the parenchymal cell clusters with sharp edges and numerous threaded ducts , occasionally having marginal pitted ducts ;its wood fibers were in bundles mostly. Results of TLC showed that the spots of the same color were found in the corresponding positions of chromatogram for 5 substance control and samples. The linear range of scopoletin, chlorogenic acid , caffeic acid , scopolactone and 3,5-dicaffeoylquinic acid were 85.60-428.00, 10.16-101.60, 10.20-102.00,40.84-408.40 and 40.80-408.00 μg/mL(all r>0.999 0). RSDs of precision ,stability,repeatability tests were all less than 3.00%(n=6). The average recoveries were 103.07%,99.66%,98.37%,97.78%,98.40%(all RSDs <3.00%,n=6). The contents of the above-mentioned 5 compounds in 10 batches of samples were 0.36%-1.23%,0.09%-0.51%,0.04%-0.13%, 0.61% -1.13% ,0.12% -1.11% ,respectively;the average com contents of water ,total ash and water soluble extract were 6.25%,5.86%,26.50%,respectively. CONCLU SIONS:O the basis of the original quality standard of A. sacrorum , microscopic identification,TLC identification ,content determination and examination items of water ,total ash and extract are added. The method shows good precision ,accuracy and stability ,which can provide reference for more scientific and standardized evaluation of the quality of this medicinal material.

ABSTRACT

Objective@#To analyze the expression and clinical significance of prohibitin (PHB) in invasive breast cancer (BC) based on high throughput multi-omics databases. @*Methods@#The breast cancer data were downloaded from TCGA and METABRIC databases, and the expressions of PHB in BC tissues and adjacent normal tissues were compared by various bioinformatics tools. The correlations of PHB expression with clinicopathological features and prognosis of BC patients were analyzed, and the interaction and function of PHB protein were predicted. @*Results@#Compared with adjacent normal tissues, PHB was highly expressed in many kinds of cancer tissues, especially in invasive breast cancer where the gene mutation and expression changes of PHB had higher proportion. The expression level of PHB had good diagnostic efficacy for BC (P<0.01). The expression level of PHB was significantly correlated with the expressions of ER and HER2, PAM50 typing and tumor purity of BC patients (P<0.05). The survival analysis showed that the high expression of PHB was an independent risk factor of BC (P<0.01). [STBX]HRAS, KSR1[STBZ] and ARAF interacted with PHB, with significant correlation. The changed expressions of [STBX]HRAS, KSR1[STBZ] and ARAF could be found in BC tissues. @*Conclusion@#The expression of PHB increases in various cancer tissues such as breast cancer, ovarian cancer and so on. The high expression of PHB has significant influence on the prognosis of BC patients. The expression of PHB has good diagnostic efficacy for BC, which may be used as a potential marker for the diagnosis and prognosis of BC.

ABSTRACT

Objective To evaluate the efficacy of case management care practice on the self-care ability of the elderly patients with prostate cancer during the endocrine therapy. Methods About 84 patients were divided into two groups by drawing lots. About 41 cases in the control group continued their treatment after discharge and consulted the nurses or doctors by phone call in case of problems. The 43 cases in the observation group was treated with case management for 3 months. The self-care ability was compared between the groups before and after the intervention. Result After the intervention, the ESCA total score of the observation group, self-care skills, self protection responsibility and health knowledge level were significantly higher than that of the observation group before intervention and that of the control group (P<0.05). Conclusion Implementation of the case management can improve the self-care ability, health knowledge level, self-care skill and sense of responsibility of the elderly patients with prostate cancer during the endocrine therapy, eventually improving the patient's ability in self care.

ABSTRACT

BACKGROUND:Rat models of complete spinal cord transection are common models for neural tissue engineering. After transecting the spinal cord by the previous methods, gap length of broken end cannot keep relatively uniform, so we cannot objectively evaluate effects of various treatments or tissue engineering materials. OBJECTIVE:The spinal cord transection models were established by using double edged micro scissors, andthe feasibility of this new model was explored by comparing with the conventional method. METHODS: A total of 42 adult female Sprague-Dawley rats were divided randomly into group A (n=6), group B (n=18) and group C (n=18). Group A only received laminectomy. In the group B, the spinal cord was transected with a sharp-pointed knife. Knife point should touch anterior wal of spinal canal and sidewal bone surface. Complete spinal cord transection models were prepared by repeated cutting. In group C, complete spinal cord transection models were established by using self-made double edged micro scissors. RESULTS AND CONCLUSION: (1) At 1 week after model establishment, in the groups B and C, complete paralysis of the hind limbs was found, and BBB scores were similar. However, significant differences in the spacing of broken end were detected. (2) At 4 weeks after model establishment, hind limb functions could restore to different degrees in both groups, but no significant difference in BBB scores was found. (3) At 8 weeks after model establishment, significant differences in hindlimb motor function scores were detectable between both groups. Biotin glucosamine tracer display: In group B, a few labeled axon fibers were observed at the caudal side of the injured spinal cord. In group C, spinal cord was completely transected, and labeled axon fibers cannot be found at the caudal side. (4) Results suggested that the modeling method of self-made double edged micro scissors could effectively eliminate individual differences, contribute to quantitative analysis and comparative study of therapeutic effects.

ABSTRACT

BACKGROUND:Vertebroplasty with bone cement injection can achieve a correction of kyphosis, enhancement of vertebral strength, and elimination of vertebral lesions during reduction of the fracture. OBJECTIVE: To analyze the efficacy of vertebroplasty with bone cement injection on osteoporotic vertebral compression fracture. METHODS:Totaly 84 patients with osteoporotic thoracolumbar vertebral compression fractures (T6-L4), 37 males and 47 females, aged 58-80 years, were randomized into two groups: study group undergoing vertebroplasty with bone cement injection and control group subject to bed rest and conservative treatment (functional exercise of the back muscle). Visual analog scale score, Oswestry disability index and vertebral height were detected and compared between the two groups before and after treatment. RESULTS AND CONCLUSION:There was no difference in vertebral height, visual analog scale score and Oswestry disability index between the two groups before treatment. At 3 months after treatment, the vertebral height was (1.653±0.168) cm in the study group and (1.521±0.200) cm in the control group, with a significant difference (P 0.05). These findings suggest that the bone cement injection as vertebral augmentation therapy can rapidly relieve pain, improve patients' quality of life within a short term and restore the vertebral height in patients with osteoporotic vertebral compression fractures.

ABSTRACT

BACKGROUND:There are reports concerning differentiation of adipose-derived mesenchymal stem cells(ADSCs)into chondrocytes using gene transfection technique.However,the transfection of adenovirus and adeno-associated virus into ADSCs is various.It is controversial whether adeno-associated virus(AAV)can transfect ADSCs.OBJECTIVE:To observe the enhanced green fluorescent protein(EGFP)expression following Ad5-EGFP and rAAV2-EGFP transfection into ADSCs,and investigate the cell proliferation ability following transfection.METHODS:ADSCs were isolated from the adipose tissue,which was from 6-month-old New Zealand albino rabbit back and neck by mechanical digestion and enzyme digestion,then ADSCs were cultured and amplified in vitro.ADSCs were infected with Ad5-EGFP and rAAV2-EGFP,and the EGFP expression was observed.A total of 2 μL sodium butyrate(1 mol/L)was added into the medium after rAAV2-EGFP transfection.MTT assay was used to detect the gene transfection effects on reproductive activity of ADSCs.RESULTS AND CONCLUSION:ADSCs isolated and cultured in vitro were flat,long-spindle and amplified stabry.The cell morphology was uniform.Many green fluorescent cells were observed in Ad5-EGFP and rAAV2-EGFP groups.Transfection efficiencies were about 88% and 83%.Adenovirus and adeno-associated virus vector can be transfected with ADSCs,and transfection efficiency is high.Adeno-associated virus needs sodium butyrate to increase its level of gene expression.

ABSTRACT

Objective To observe the effects of preoperative acute hypervolemic hemodilution (AHH) on coagulation and fibrinolysis on the elderly patients undergoing hepatic carcinectomy.Methods Thirty ASA Ⅰ~Ⅱ liver cancer patients over sixty years old undergoing scheduled elective hepatic carcinectomy were random divided into AHH group ( n = 15) and control group ( n = 15).AHH was performed by infusing 15 ~20ml/kg 6% Voluven ( 130/0.4) in a rate of 30ml/min on patients in AHH group,and the patients in control group were infused routinely lactated Ringer's solution.Blood samples were collected for measuring coagulation function and platelet activation before anesthesia (T1) ,30 minutes after AHH (T2),1 hour ( T3 ), immediately (T4) and 24 hours (T5) after operation in group AHH, and the similar procure was used in control group.Results There was significant difference in the volume of allergenic blood transfusions between the two groups(20% vs 53.3%, P <0.05).PT and APTT in both groups prolonged significantly after T2, but they all remained in normal range.TT and DD had no significant changes between the two groups.SFMC and F1 + 2 increased in both groups, but it had no significant changes.PAC-1 and CD62P expressions of group AHH were significantly lower T2, T3 ,respectively ( 1.37 ±0.4) %, (1.28 ±0.4) %, than those at T1 [( 1.96 ± 0.6) %] ( P < 0.05 ).Compared with control group, CD62P was significantly lower in AHH group at T2 and T5.Conclusion It is safe to take AHH of Hct 25% to 30% during hepatic carcinectomy for elderly liver cancer patients without cardiac and pulmonary disease.And it could decrease allergenic blood transfusions, with no apparently influence on coagulation function.

ABSTRACT

OBJECTIVE: To optimize the extraction technology of thrombolytic components predominantly as corilagin from Phyllanthus urinaria.METHODS: The extraction technology was optimized by orthogonal experiment with solvent amount,extracting time and extracting times as factors,and with the comprehensive score of the yield of extract and corilagin content as indexe.The impurity-removing efficacy by centrifugation,acid/base method or ethanol precipitation was evaluated.RESULTS: The optimal extraction technology was as follows: using 8-fold volume of 30% ethanol as solvent for reflux extraction of 3 times(2 hours/ time).The best impurity-removing efficacy was achieved when 90% ethanol was used for precipitation.CONCLUSION: The extraction technology is simple and stable with high yield of active components,and it provides theoretic basis for the industrial production.

ABSTRACT

We aimed to define the detailed clinical features of Japanese childhood-onset Type 2 diabetes mellitus [T2DM] patients and to determine whether their discernable characteristics were similar to those of adult- and childhood-onset T2DM in other countries. Research design and methods: Subjects were 22 patients [10 males and 12 females] under treatment without hepatocyte nuclear factor-1alpha or mitochondrial gene mutations and who were diagnosed as diabetic when less than 15 years of age. Results: Body mass indices in boys and girls at onset were 25.8 +/- 6.3 and 24.7 +/- 3.6, respectively, with mean ages of 13.3 +/- 1.7 and 12.8 +/- 2.0 years, respectively. Most patients had a short diabetic duration that required insulin treatment. One or both parents of 18 of the 22 T2DM subjects were diabetic and seven subjects had a history of diabetes in their family over three generations. Conclusion: We demonstrated that a relatively high number of Japanese childhood-onset T2DM cases have a strong genetic factor, and are not necessarily related to excessive obesity. Furthermore, most patients required insulin therapy in the initial stages because of insufficient pancreatic beta-cell reserves. This suggests that malfunction of pancreatic beta-cells triggers hyperglycemia resulting in the requirement for insulin in some Japanese childhood-onset T2DM patients

ABSTRACT

OBJECTIVE: To optimize the extraction conditions of plumula nelumbinis. METHODS: The semi - bionic extraction (SBE) conditions were optimized by homogeneous design while total alkaloids and dry extract were adopted as markers. Factors of extraction time, solute quantity and the pH values were evaluated for their effects on extraction process. RESULTS: Optimal parameters were as follows: plumula nelumbinis was extracted in water 8 times as much as it for 3 times; the pH values of the water for the 1st, 2nd and 3rd decoctions were 5.0,6.0 and 8.0, respectively; the boiling lasted 4 hours. CONCLUSION: The method prouides theoretical reference for the extraction plumula nelumbinis.

ABSTRACT

【Objective】To optimize the functional monomers which is used for the synthesis of Corilagin molecular imprinting polymer.【Methods】The chromatographic characteristics of Corilagin molecular imprinting polymers synthesized respectively by functional monomers of ?-methacrylic acid(MAA)and acrylamied(AA)were investigated with HPLC,and the imprinting efficiency of the two kinds of polymers on Colilagin template were compared.【Results】The polymer with AA as the functional monomer had a higher imprinting efficiency than the polymer with MAA as the functional monomer.【Conclusion】AA is the optimal functional monomer for the synthesis of Corilagin molecular imprinting polymer.

OBJECTIVE:To optimize the extract technology for Plumula Nelumbinis.METHODS:Four methods:water extraction(WE),water extraction by precipitation with alcohol(WAE),semi-bionic extraction(SBE),and semi-bionic ex-traction by precipitation with alcohol(SBAE)were used to extract Plumula Nelumbinis and also compared in their effects by adopting the total content of alkaloid and the yield of dry extract as markers.The comprehensive values Y was calculated.RESULTS:Through the comprehensive evaluation of the two markers,the comprehensive values Y ranked in the order of SBE,WE,SBAE and WAE.CONCLUSION:SBE method is the optimal method to extract Plumula Nelumbinis.

ABSTRACT

Objective To compare the effect of ultrafiltration method and activated carbon absorbance method on aconitine-type alkaloid (AA) content in the preparation of Yiqi Huiyang Injection (YHJ) and to evaluate the safety of YHJ by limited detrmination of diester alkaloids (DEA).Methods Acid dye colorimetry was used to detect AA content and reverse phase high performance liquid chromatography (RP-HPLC) was applied to determine the limited content of DEA. Mesaconitine, aconitine, hypaconitine were separated on a Hypersil BDS-C?18?column, with methanol and 0.05% triethylamine (70∶30) as mobile phase, wavelength at 240 nm and external standard for quantitative analysis.Results Compared with activated carbon absorbance method, the loss of AA was reduced, limited content of aconitine was within the pharmacopoeia standard method and the other two kinds of DEA were also low in YHJ prepared by ultrafiltration method.Conclusion Ultrafiltration was superior to activated carbon absorbance in the preparation of YHJ as with AA as the parameter, and the contents of DEA were very low which indicates that the toxicity of YHJ may be also low.

ABSTRACT

Objective To study the safety of Plumula nelumbinis microcapsules and its anti-arrhythmic effect.Methods Acute toxicity test in mice,arrhythmia mice models induced by chloroform and arrhythmia rats models induced by barium chloride(BaCl2)were adopted.Results The maximum tolerance dose for mice was 16.0 g/kg body weight.The microcapsule decreased the incidence of chloroform-induced ventricular fibrillation in mice significantly(P 0.05,compared with the blank control group).Conclusion The preliminary results show that Plumula nelumbinis microcapsule is safe,and exerts anti-arrhythmic effect against experimental arrhythmia.

Objective: To study various preparation methods for Tongmai Oral Liquid. Methods: Tongmai Oral Liquid with ultrafiltration method, water alcohol method, talcum powder method, and method were prepared respectively; using impurity clearing efficency, clarity, total saponin content as parameters for quality control. Results: Tongmai Oral Liquid prepared by water alcohol method (alcohol concentration being 75%) was better than that by other methods in claerity, and stability and has highest total saponin. Conclusions: Quality of Tongmai Oral Liquid prepared by water alcohol method was available.

Objective To analyze the changes of insulin-like growth factor-1(IGF-1) serum level in diabetic lower extremity angiopathy,explore the role of IGF-1 in the pathogenesis of diabetic vascular complications,and provide basis for prevention and treatment of diabetic angiopathy.Methods We selected 74 type 2 diabetes mellitus(T2DM) patients as case group and 15 normal people as control group.All of the research subjects were measured in height,weight,blood pressure,blood glucose,blood lipid,fast insulin,and urinary albumin excretion rate(UAER).The serum IGF-1 level was measured with ELISA.Arteries of both lower extremities were measured with color Doppler flow image.Results ① The serum IGF-1 level was significantly higher in DM patients than in control group.② The serum IGF-1 level was significantly higher in diabetic patients with lower extremities arterial vasculopathy than diabetic patients without.The results showed a positive correlation between the serum IGF-1 level and severity of lower extremity arterial vasculopathy.③ There was a positive correlation between diabetic duration,UAER,total cholesterol(TC),lower density lipoprotein(LDL) in blood and lower extremity arterial vasculopathy in diabetic patients.Conclusion The serum IGF-1 level can reflect the severity of lower extremity arterial vasculopathy.The serum IGF-1 level may be a reference valuable in diagnosis of diabetic macroangiopathy and a new cure point for diabetic macroangiopathy.
